Gambir is one of the agricultural commodities with a high economic value and has many benefits that are used as raw materials for various industries. Utilization of gambier to produce secondary metabolites can be done by vegetative and generative propagation, but requires a long production time and has limited raw materials and the quality of the results is not uniform and the low quality produced. In vitro culture is one alternative that can be done to obtain secondary metabolites in a relatively short time by inducing callus. This study aims to obtain the optimal concentration of 2.4-D, kinetin and their interaction in inducing callus. This study used a 2 factor Randomized Group Design with 12 treatments and 3 groups. The first factor is 2.4-D which consists of four concentration levels: 0 ppm; 1 ppm; 2 ppm; 3 ppm) and the second factor is kinetin which consists of three concentration levels: 0 ppm; 0.5 ppm; 1 ppm). The observed parameters included time to callus appearance, percentage of callus explants, callus texture and color. The results showed that the average 2.4-D 3 ppm was able to accelerate callus induction with a time of 7.66 DAP with a callus color of 33.33% yellowish white and 33.33% compact texture and the interaction of 1 ppm 2.4-D and 1 ppm kinetin was optimal in inducing callus of gambier plants when callus appeared 9.72 DAP, callus color 61.11% yellowish white and callus texture produced 72.22% compact.
